Abstract:

     Lipoxygenase(LOX) is a key enzyme in jasmonic acid (JA) biosynthesis pathway. In this study, a full-length cDNA clone of LOX  (HbLOX1) and its promoter were cloned from rubber tree (Heveabrasiliensis). Nucleotide sequence analysis revealed that the cDNA clone (HbLOX1)was 3,031 bp encoding for a polypeptide of 906 amino acids with molecular weight of 102.59 kDa and pI of 5.68. The deduced amino acid sequence showed N-terminal chloroplast transit peptide and classified as 13-LOX. Real-time PCR analysis showed that HbLOX1 gene was progressively up-regulated by ethylene, the hormone usedas stimulant to increase the latex yield. In addition, the HbLOX1 promoter showed many cis-acting regulatory regions responsive to plant hormones including ethylene and methyl jasmonate (MeJA). These confi rmed the well-known crosstalk between the ethylene and JA pathway in plants.According to the importance of JA pathway in the regulation of gene expression cascade that may lead to increase in latex yield, the expression of other genes involved in JA biosynthesis should be investigated.
